Mushroom DNA isolation at home

Whenever we think about DNA, We consider it is a complex structure and it's extraction is a complex process and it is possible at the high-tech laboratory but today my blog will tell you how to isolate DNA at home with a household material in a very easy manner ๐Ÿ˜Š

But what is DNA๐Ÿค”๐Ÿ˜‘ - It is defined as Deoxyribonucleic acid,  a double-helical structure and consist of genetic information because of DNA, mushrooms that we consume today gave  the same structure and taste that our grandfathers had.
I hope I have cleared your doubt about DNA.

Let's start our isolation process 

Principle
DNA of  mushrooms is a  eukaryotic cells.To isolate DNA we have to break down the cell wall, cell membrane, and nuclear membrane to release the DNA. The grinding of the sample destroys the cell wall and cell membrane. After the addition of NaCl, it removes the protein and the protein gets dissolved in an aqueous layer so they don't precipitate in the alcohol along with DNA most importantly, sodium ions create a temporary bond between positive sodium ion and negative phosphate backbone and make a stable DNA, thus making DNA more hydrophobic. Detergent plays a major role in breaking the cell membrane and nuclear pore, making it exposer DNA ( DNA is present inside the nucleus), and helping in the removal of lipid molecules, pineapple juice consists of bromelain enzyme and this enzyme helps in breakdown protein bound to DNA and at last, ethanol helps in the precipitation of DNA.

Material Required 
NaCl (common salt), washing powder, freshly prepared ( pineapple juice), chilled 70% ethanol (rubbing alcohol), chilled water, sample - mushroom, Grinder or mortar and pestle, stained, beaker and test tube 










Procedure 
1. Weight 50gms of mushroom sample and add 100 ml of chilled water, put a half tablespoon of NaCl, and grind it in a jar. 
2. Filter the blended mixture into a beaker using a stainer. Add 1 tablespoon of detergent to the filtered mixture and stir it properly.
3. Wait for 10 minutes and transfer the mixture to a test tube and add 5ml of freshly prepared pineapple juice.
4. Wait for 5 to 10 minutes and gently shake the test tube 
5. Hold the test tube in an inclined position and gently add 5ml of chilled 70 % chilled ethanol with the help dropper.
6. Observe the two phases between the mixture and ethanol you will oobservea aread-like precipitate in the above phase. You can spool the DNA with the help of a needle or a thin glass rod  
It's all  about the extraction of DNA.
